This research studied the in vitro compatibility of Cherax quadricarinatus digestive proteases and crude enzyme extracts recovered from fish‐processing waste to evaluate their potential use as feed additives to increase digestion efficiency in decapod crustaceans. The sources of exogenous enzymes were: Pleoticus muelleri (Pm), Artemesia longinaris (Al), and Illex argentinus (Ia). The midgut glands of early (7.2 ± 0.34 g) and advanced juveniles (13.8 ± 1.49 g) of C. quadricarinatus were used for all the assays. The compatibility and synergy between crayfish and exogenous enzymes were evaluated through sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS–PAGE) electrophoresis and the hydrolysis of azocasein and four different meals. SDS–PAGE gels demonstrated that crayfish bands remained active when their enzymes were combined with the enzymes from Pm and Ia. An apparent enzymatic synergism was found when Pm and crayfish proteases were incubated together at pH 6 and 8 with azocasein (p ? 0.05). The ability to hydrolyze the different feed ingredients varied among the enzyme combinations assayed. Our findings suggest that fish waste has potential as a source of proteases, adding value to these processing residues and promoting a zero‐waste industry. Enzymes obtained from Pm and Ia are suitable to be used as food additives for crayfish. The results should be validated through feeding trials.  相似文献

The optimization of reproductive parameters in shrimp farming continues to be a challenge for most producing countries. Although the crustacean neuropeptides have been studied extensively in the last two decades, the functions of most of these neuropeptides remained putative. Among them, molt‐inhibiting hormone isoform II (MIH II) has shown an important role in vitellogenesis. In this study, the cDNA encoding mature MIH II peptide was isolated by RT‐PCR from the L. vannamei eyestalk. The cDNA was cloned into pET28a bacterial expression vector. Recombinant MIH II was obtained in the form of insoluble inclusion bodies and purified to ~88% purity. Two doses of rMIH II and a negative control group were assayed in vivo. The stages of ovarian maturation and spawning were recorded during 72 hr post‐injection. The results showed that ovarian maturation occurred approximately in 9% and 33% of females injected with rMIH II at the doses of 300 and 600 ng/gbw respectively. Neither maturation nor spawning was detected in the negative control group. Females injected with 600 ng/gbw, which showed vitellogenic stages III and IV, spawned. These preliminary results argue that the hormone rMIH II could be a promising candidate to induce spawning in L. vannamei shrimp.  相似文献

In Venezuelan Amazonian, some producers have established small agroforestry systems of production on sandy savanna soils by a long-term addition (more than 25 years) of animal manures at a low dressing (2 Mg ha−1) as fertilizer input. As a result of the organic fertilizers regime, the original savanna soil has been changed in terms of soil quality parameters. The main objective of the study was to investigate using sequential fractionation of soil P the impact of organic manures on the amount and partitioning of bioavailable P in soils of the Amazonas. Fractionation was carried out on Typic Ustipsamments amended with three different organic manure sources for extended periods. In general, after fertilization, all Pi and Po fractions increased significantly. The increase was striking in the resin-Pi and HCl-Pi, and among the organic P fractions, the changes were highest for the NaOH-Po sonicated and non-sonicated fractions. The total P increment was more relevant when soils were amended with chicken manure (1,194 mg Pt kg−1) and less relevant for the farm soil treated with compost (500 mg Pt kg−1), where the soil amended with cattle waste presented an intermediate value in total soil P (851 mg Pt kg−1). The importance of this field study was to assess the sustainability of long-term established organic management characterized by the low inputs, and this information is poor in the Amazonas.  相似文献
